Key scientific and processing issues faced by the pharmaceutical industries involve control of the solid-state form production of active pharmaceutical ingredients (APIs). Most drugs available on the market are commercialized as solid forms, and the particular form produced, or even the size of the crystalline particles, may affect parameters of paramount importance for the patient, as solubility, bioavailability, stability towards temperature and humidity, and even taste. Co-crystals (two or more molecules combined in the same crystal) and polymorphs (only one type of molecule resulting in different crystals/architectures) are emerging as an important technology, allowing the modification of solubility and bioavailability of target APIs, nutraceuticals, and other agents through interactions with appropriately selected co-formers (different molecules). The great potential of co-crystals resides in their different properties with respect to those of the single entity crystals; at the same time the process of co-crystal formation does not alter the chemical integrity of the component of interest. The high potential of co-crystals to fine-tune properties for a given API is becoming clear and can now be adapted across a broad range of drug molecules. The motivation for many researchers in this field (systematic commercialization of co-crystal drug products with advantageous clinical safety and efficacy) has coincided with development of the intellectual property landscape.
